| 内容記述 | Human population growth has increased the demand for human necessities such as food, clean water, shelter and energy. As a result, the number of wastewater treatment plants (WWTPs) around the world has increased to accommodate the rising urbanization of human activities. Most WWTPs use activated sludge for the biological treatment to remove organic materials from wastewater. Consequently, cause an increase in the quantity of waste activated sludge (WAS), which is ultimately destined to be burned and disposed of in a landfill. In this work, WAS will be utilized as a bioenergy source by increasing methane production via quorum sensing and quenching of microbial communities. The quenching autoinducer 2 (AI-2) can obstruct intra- and interspecies microbial communication by incubating WAS with microencapsulated genetically deleted LuxS and LsrR gene double mutant E. coli. The organic acid and significant organism shift brought on by this disruption that contributed to the increase in methane have been found and highlighted in this study. Unfortunately, bioenergy generation is still not commonly practiced by WWTPs around the world, thus the vast majority of WAS will continue to be disposed of in landfills, resulting in waste greenhouse gas emissions that contribute to climate change. This study also explores the potential for WAS bioremediation using bacteriophage. Isolated bacteriophages capable of lysing bacteria with antimicrobial resistance (AMR) were identified by using whole genome next generation sequencing technology. The bacteriophages were then mixed to form a cocktail phage (CP), which was then cultured with WAS. This anaerobic phage therapy resulted in considerable reductions in harmful hydrogen sulfide gas and total methane inhibition. Methanogen and sulfur-reducing bacteria imbalance caused by phage therapy are regarded to be the key organisms in this research. | |||||||
